Yamamoto started Game 1 against the San Diego Padres in the 2024 Major League National League Division Series (DS) at Dodger Stadium in Los Angeles, California on the 6th (Korea time), but gave up three runs, allowing one home run in the top of the first inning.

Yamamoto received a left-handed hit by Luis Aras, the first batter, and faced Fernando Tatis Jr., the subsequent batter, and was on the verge of losing a point due to the catcher's foil. Yamamoto, unable to concentrate on the match against the batter, allowed a walk after wild pitches, and grounded out Jurickson Profar at the first and third bases, but in the meantime, he failed to stop Aras from scoring. Yamamoto was hit by a splitter in the ensuing match against Sandy and leading hitter Manny Machado, and he even hit a two-run homer to the left. He gave up three runs in an instant. Yamamoto, who was shaken after a hit by Gender Bogaerts, caught Jake Cronenworth with a ground ball to finish the first inning in 28 pitches.

Yamamoto signed a 12-year contract for 325 million dollars and about 433.3 billion won before this season, surpassing the ransom of Garrett Cole, the New York Yankees who won the previous MLB pitcher's best contract. In this season's regular season, he pitched in 18 games with 7 wins and 2 losses and an ERA of 3.00. Due to his injury, he has been undergoing rehabilitation treatment for more than two months since mid-June, and has faced this postseason after returning to the stage at the end of the regular season.

Manager Dave Roberts originally announced Jack Flaherty as the starting pitcher for Game 1, but replaced him with Yamamoto. The Dodgers was in an unbecoming situation for its reputation as a "pitcher kingdom" as its main starting pitchers have been sidelined one after another due to injuries. Yamamoto, regarded as one of the best in the league in terms of potential, took on a heavy task in Game 1, but gave up the momentum. Yamamoto hit all three batters he faced starting with the eighth batter in the second inning.
